I sampled 'High Peak's 58 service from Macclesfield - Buxton yesterday and was not disappointed. It travels on the A537 from Macclesfield to climb almost 1200 feet from Macclesfield Bus Station at about 450 feet ASL to reach 1690 feet (515m for the younger readers). There are sweeping sharp bends and wide open vistas over the surrounding countryside. The service runs hourly and I would recommend the journey. I think this route must reach the highest altitude for a regular service (ie not Summer or our own 351 over Holme Moss service to Glossop run by South Pennine on Saturdays) - maybe somebody out there with a similar interest to me on extremes may think of one!

not sure if this would count as it mostly uses a branded coach rather than buses these days, but in Scotland route 37/X37 from Inverness & Aviemore to the Cairngorms reaches just over 2000 feet at the Cairngorms mountain car park where it terminates.

Not quite as high as those, but I think the highest in our area is the B3 (432m, 1,418ft).

New contender (for West Yorks). Greenfield Road between Holmfirth and Greenfield tops out at 499m. I believe the 352 and 357 go up there.

Some interesting comments - there is also the Summer only Wrightbus 888 Service from Newcastle - Keswick over the A586 Hartside Road which reaches 580m. I believe the highest ever (possible) bus route in the UK was an extension of the spectacular Stagecoach 201 service from Aberdeen to Braemar and then over the highest A road in the UK (A93) to reach 665m to Blairgowrie. I think not too surprisingly, it only ran for one Summer Season!
